Dysfunctional glucocorticoid receptors in acute leukemia.

چکیده
The obligatory role of specific cytoplasmic receptors as mediators of steroid hormone action is well established. In lymphoid cell culture Systems the development or glucocorticoid resistance is almost always due to a mutation to a receptor negative state [I]. Therefore, if the assumption is that glucocorticoid-induced remission in leukemia is by a direct effect on leukemic blast cells, one would predict that the presence of absence of blast cell glucocorticoid receptors would be a major determinant or clinical steroid sensitivity. However, receptor quantization has not correlated with steroid responsiveness [2-51, and in particular, high receptor numbers are not uniformly associated with responsive disease [6] . We have speculated that this lack of concordance between receptor Status and clinical outcome may be due to the presence of glucocorticoid binding macromolecules which are physiologically nonfunctional in those leukemias which are glucocorticoid resistant. To test this hypothesis, we have studied certain biochemical and biophysical characteristics of glucocorticoid binders in normal and leukemia cells, to determine if such dysfunctional binding macromolecules exist, and to establish their relationship to therapeutic outcome. Glucocorticoid receptors and in vitro responses to glucocorticoid in acute nonlymphocytic leukemia.
Early clinical studies in which glucocorticoids were used alone in the treatment of acute nonlymphocytic leukemia (ANLL) reported a wide range of responses from remission in some patients to dramatic aggravation of the disease in others. In the hopes of identifying those patients likely to derive therapeutic benefit from glucocorticoids, we have studied glucocorticoid receptors and in vitro res...

Background: Previous studies suggested that stressful events that release Glucocorticoid from adrenal cortex and also injection of agonists of glucocorticoids receptors probably affect emotional learning and memory process and modulate them. The aim of this study was to determine the effects of acute stress and systemic injection of Corticosterone (as agonist of glucocorticoid receptors) on acq...
متن کاملGlucocorticoid sensitivity and receptors in cells of human myelogenous leukemia lines.
The effect of dexamethasone on the clonal proliferation of human acute myelogenous leukemia cells was studied using the two available human acute myelogenous leukemia lines KG-1 and HL-60. The cells were cloned in agar in the presence of colony-stimulating activity with and without dexamethasone. Colony formation by KG-1 cells was inhibited by dexamethasone at a concentration of 10(-7) M, and a...
متن کاملClinical implications of glucocorticoid receptor studies in childhood acute lymphoblastic leukemia.
We have performed in parallel, in 19 children with acute lymphoblastic leukemia, a quantitative determination of glucocorticoid levels, in vitro steroid induced inhibition of nucleic acid precursors, and a short-term clinical trial of corticosteroids alone, before the treatment was given, which included corticosteroids and other drugs. From our results it appears that high glucocorticoid recept...
